First you need to fetch file path from Database. Once you get the file path you need to delete it. For getting the file path from DB you can use ADO.NET or EntityFramework.

Below code is to delete file from Server:

string path = GetFromDB(); // Implement your logic
if(File.Exists(path))
{
File.Delete(path);
}

Once you delete file successfully, update your DB column as blank value for particular record.
